In the U.S. there are 17 million people with Major Depressive Disorder (MDD). As many as 30-40% of those people have a treatment-resistant form of the disorder (TRD).  Nearly one half of those diagnosed with depression also have anxiety disorder and vice versa. Ketamine is a dissociative anesthetic that has been used safely and extensively since FDA approval in 1970.  Ketamine is also included on the WHO list of essential medications, due to its wide margin of safety across all age groups.

Ketamine works by binding to N-methyl-D-aspartate receptors causing a disinhibition pyramidal cells which cause increased  Glutamate release.  Multiple lines of research evidence also show that BDNF (brain derived nerve factor) serves a critical role in the pathogenesis of depression and ketamine’s rapid-acting effects are through the rapid BDNF synthesis pathway.  Through functional imaging scans, ketamine appears to aid with depression by activating the anterior cingulate cortex and increasing connectivity between the insult and default mode network.
